Cardiolipin antibody and lupus anticoagulant is used along with the tests of Beta-2 glycoprotein 1 antibody to help detect the cause of the thrombotic episode or recurrent miscarriages caused due to unexplained blood clot. It is done if a blood clot is suspected in a vein or artery. Short of breath, pain in the upper and lower limb extremities and headache are the common symptoms when the above mentioned test is recommended. APS, Antiphospholipid Syndrome, is a disorder that contains symptoms like thrombosis of veins and arteries, ulcers of the lower limb, loss of fetus during pregnancy, thrombocytopenia, haemolytic anaemia, etc.

For the test to be carried out, preferred sample matrixes are human serum or plasma. The sample needs to be pre-diluted using the diluent in the ratio of 1:100. For e.g., 10 μL of sample should be diluted with 990 μL of sample diluent. Blood is collected from the patient through the venipuncture method in the vacutainers and serum is separated from the blood after the formation of the clots or by the method of centrifugation. Bilirubin and haemolysis isn’t required for this test to be carried out. The sample collected might be stored at -20°C for up to six months.

All the reagents to reach the room temperature level by letting them sit for at least 30 minutes. In order to avoid the microbial infection, the unused reagents must not be transferred to the original vessels. Incubate the sample for at least 60 mins at room temperature. Then remove the content from each well and wash the wells with 300 μL of diluted wash solution at least 3 times. Again incubate the sample left for 30 mins at room temperature. Repeating the same procedure, remove the content from the well and wash the well 3 times with 300 μL of diluted wash solution. Incubate the obtained TMB substrate for 15 minutes in the dark at room temperature. Finally, shake the microplate gently and note the results by making calibration curve.
