I am taking some medicine for bulge spines and Lumber spondylitis, and my urine passes little painful and colour of urine became little reddish. But if I drink 2-3 glass of water at a time, it becomes of natural colour. Should I test my urine sample?

The medicines that you mentioned should not change the colour of urine when taken. As you explained it appears to be simple dehydration. But if you have complaints of burning sensation during urination then you should definitely do your urine test.
