Rosai-Dorfman disease is a rare disorder characterized by overproduction and accumulation of a specific type of white blood cell in the lymph nodes. The most challenging adverse effect of thalidomide is neuropathy, which might be persistent at a defined cumulative dose. Steroid therapy profoundly inhibits lymphocyte migration into lymph nodes thereby disrupting lymphocyte recirculation. Thalidomide must not be taken by women who are pregnant or who could become pregnant while taking this medication. Even a single dose of thalidomide taken during pregnancy can cause severe birth defects.
